Research Project: OPTIMIZATION OF SAMPLE PREPARATION FOR ANALYSES OF PHENOLIC COMPOUNDS FROM DIFFERENT MATRICES

2010 Annual Report


1a.Objectives (from AD-416)
Optimization of sample preparation procedures for extraction of phenolic compounds from different food and plant matrices.


1b.Approach (from AD-416)
The approach will include samples to be extracted by different procedures such as Stirring, Sonication, Shaking and Accelerated Solvent Extractor. Impact of solvent, temperature, and particle size on extraction of phenolic compounds from different food matrices will be evaluated.


3.Progress Report

This is a continuation of a previous agreement with Dionex. Dionex has furnished FCMDL with parts, maintenance, and technical support for the accelerated solvent extractor (ASE) in exchange for research on the extraction of lipids and phytochemicals and presentations at technical meetings describing the use of the ASE. The ASE is used routinely at FCMDL to compare and develop methods that provide the maximum yield for phytochemicals. It allows optimization of extraction solvent, time, temperature, repeat cycles, solvent-solid ratios, rinse solutions, and multiple solvents. In this fiscal year, it has been applied to the extraction of lipids and isoflavones from soy, artemisinin and flavonoids from Artemisia, flavonoids from wheat fractions, phenolic acids from potato skins, and volatile aromatic compounds from citronella and lemon grasses.

This agreement was monitored through routine conversations and emails with the Dionex Corporation.
